Crank Mechanism

Name: Monish Kumar (S11065194) The University of the South Pacific MM313 Dynamic Systems Experiment 2- Crank Mechanism Aim: To investigate the relationship between piston displacement and crank angle for different ratios between the connecting rod and the crank. Also to look at the relationship between the turning moment on the crank shaft and crank angle for a given force on the piston. Equipment and Instrument: Introduction: A crank is an arm attached at right angles to a rotating shaft by which reciprocating motion is imparted to or received from the shaft. It is used to convert circular motion into reciprocating motion, or vice-versa.The arm may be a bent portion of the shaft, or a separate arm attached to it. Attached to the end of the crank by a pivot is a rod, usually called a connecting rod. The end of the rod attached to the crank moves in a circular motion, while the other end is usually constrained to move in a linear sliding motion. Theory: Figure 1. 0: Slider crank mecha nism The slider crank mechanism as shown in figure 1. 0 is a kinematic mechanism. The piston displacement from the top dead centre, x, can be determined from the geometry of the mechanism, in terms of the lengths of the connecting rod, L, and crank, R, and the crank angle, ? can be expressed as x=L+R-(Lcos? -Rcos? ) Also from the geometry, it can be seen that Rsin? =Lsin? And sin? =sin? n Hence cos? =[1+sin? n2]1/2 Where n is a ratio: n=LR Procedure: Part A: 1) No weights and hangers required, the unit initial starting position 0 in the protractor is setup and 90? and 270? protractor positions to be in line with the level lines in each side. 2) The unit is to be setup in its highest point, Top dead centre point was used to work out the displacement value 3) The mounted disc was turned 30? nd the displacement was noted on the results table, this step was again repeated for different angles and different crank positions. After plotting the graph of Displacement versus the crank angle position, the graph show that the experimental values and the theoretical displacement can be compared, the experimental plot and the theoretical plot are almost same. 2. From the results graph the graph show that the measured displacement follows the theoretical curve very well. The maximum difference between the experimental and theoretical displacement is 2 mm. 3. For full rotation i. e. 60? the motion of the piston is close to simple harmonic, after 180? the displacement will gradually decrease to 0, it will form a cosine graph. The weight is 5. 8 N at LHS whereas for 4N added piston weight the greatest amount of force is 6. 5 N at 225? RHS. Conclusion:The kinematic motion of the crank mechanism can be expressed in terms of the lengths of the crank and the conrod, and the displacement of the crankshaft. The experimental measurements of piston displacement agree with the prediction of a theoretical model of the piston motion. Due to friction errors were made in the second part of the experiment but still manage to get the results to find out the greatest amount of force being exerted on crank mechanism. Chapter Seven The List While Jody showered, Tommy made a list. Feed Laundry New Apartment Toothpaste Sweet Monkey Love Windex Dispose of Vampire Minion â€Å"What do we need an onion for?† Jody asked. She was having a little trouble getting her vision to focus. â€Å"Minion, minion,† Tommy said. â€Å"Mint-flavored onion? Why do we need that?† â€Å"A minion! Someone who can move around during the day who can help us out. Like I was for you.† â€Å"Oh, my bitch.† Tommy dropped his list. â€Å"Nuh-uh.† Jody picked it up and walked over to the kitchen counter where the coffee machine stood. â€Å"I would sell my soul for a big cup of joe.† â€Å"I was not your bitch,† Tommy said. â€Å"Right, right, right. Whatever. So how long do we have to do this list?† â€Å"I checked the almanac. Sunrise is at six fifty-three, so we have about twelve hours. It's almost the solstice, so we get a lot of darkness.† â€Å"Solstice? Oh my God, it's almost Christmas.† â€Å"So?† â€Å"Hello? Shopping?† â€Å"Hello? We have an excuse. We're dead.† â€Å"My mother doesn't know that. I have to find something for her that she'll disapprove of. And your family – â€Å" â€Å"Oh my God! Christmas. I was supposed to go home to Indiana for Christmas. We need to redo the list.† â€Å"You do it. I'm going to dry my hair,† Jody said. The new list read: Christmas Presents Call Home Feed Minion (not our Bitch) Hot Monkey Love Windex Write Literature Dispose of Creepy Old Vampire New Apartment Laundry Toothpaste â€Å"I think you should take monkey love off of the list,† Jody said. â€Å"What if we lose the list and someone finds it?† â€Å"Well I think ‘dispose of Creepy Old Vampire' would be a little more embarrassing, don't you?† â€Å"You're right, cut monkey love and change ‘vampire' to ‘Elijah. † Jody tapped the list with a pen. â€Å"And take off Windex and put in ‘buy coffee. â€Å" â€Å"We can't drink coffee.† â€Å"We can smell it. Tommy, I desperately need coffee. It's like the blood hunger, only, you know, more civilized.† â€Å"Speaking of blood hunger – â€Å" â€Å"Yeah, you'd better move that up the list.† â€Å"And add a bottle of whiskey. You're going to have to buy it.† â€Å"Sorry, writer boy, but we're doing this stupid list together.† â€Å"I'm not old enough to buy liquor.† Jody stepped away from him and shuddered. â€Å"That's right. Isn't it?† â€Å"Yep,† Tommy said, nodding – trying to look wide-eyed and innocent. â€Å"Well, okay then. I should have checked IDs before picking my bitch.† â€Å"Hey!† â€Å"Kidding. What are you going to do with a bottle of whiskey anyway?† â€Å"Check something else off the list,† Tommy said. â€Å"I have an idea. Get your purse.† â€Å"What did the Animals want, anyway?† â€Å"Twenty grand.† â€Å"I hope you told them to fuck themselves.† â€Å"They did that already.† â€Å"Did they suspect, you know, about what you are now?† â€Å"Not yet. Lash said I looked a little pale. I sent them to the store. If Clint knows, well – â€Å" â€Å"Oh, good move. Maybe we should just take out an ad. ‘Young vampire couple seeks angry village people to hunt them down and kill them. ; â€Å"Ha. Village people. Funny. Put self-tanning lotion on the list. I think the pale thing is giving me away.† At seven in the evening, three days before Christmas, Union Square was awash in shoppers. There was a Santa's Village set up in the raised square, with a line of children and parents that wound five hundred deep through a labyrinth of red velvet cattle gates. Around the square, the street performers, who would normally have knocked off around five, lined the granite steps up to the square. A juggler here, a sleight-of-hand guy there, a half-dozen â€Å"robots† – people painted silver and gold who would move in machine-jerk rhythm for the drop of a coin or a bill – and even a couple of human statues. Jody's favorite was a gold guy in a business suit, who stood motionless for hours on end, as if he'd been frozen in midstep on the way to work. There was a small hole in his briefcase into which people stuffed bills and dropped coins after photographing him or trying to make him flinch. â€Å"This guy used to freak me out,† Tommy whispered. â€Å"But now I can see him breathing and the aura thing.† â€Å"I watched him for a whole lunch hour one time and he never moved,† Jody said. â€Å"In the summer, you know he has to be suffering in that painted suit.† Suddenly she shuddered at the thought of Elijah, the old vampire, still encased in bronze back at the loft. Yes, he had killed her, technically, but in a way he'd just opened a door for her, a door that, no matter how bizarre, was immediate, vital, and passionate. And yes, he'd done it for his amusement, he'd said, but also because he was lonely. She wound her arm into Tommy's and kissed him on the cheek. â€Å"What was that for?† â€Å"Because you're here,† she said. â€Å"What's first on the list?† â€Å"Christmas presents.† â€Å"Skip down.† â€Å"Sweet monkey love.† â€Å"Yeah, we'll do it in the Santa's Workshop window at Macy's.† â€Å"Really?† â€Å"No, not really.† â€Å"Okay, then we need liquor.† Jody snatched the list out of his hand so quickly that most people wouldn't have even seen her move. â€Å"You are no longer in charge of the list. We're getting me a new leather jacket.† I AM HOMELESS AND SOMEONE SHAVED MY HUGE CAT. William had changed his sign. Chet the huge cat was still wearing Jody's sweater. He eyed the two vampires suspiciously as they approached. Tommy held the bottle of Johnny Walker out to William. â€Å"Merry Christmas.† William took the liquor and squirreled it away in his coat. â€Å"Most people just give money,† he said. â€Å"We're cutting out the middleman,† Jody said. â€Å"How are you feeling today?† â€Å"Great, why? Really good, you know, considering that I'm homeless and you guys shaved my cat.† â€Å"You were pretty hammered last night.† â€Å"Yeah, but I feel great today.† â€Å"That's how it used to affect me,† Tommy said. â€Å"Remember. Kind of energizing.† Jody waved Tommy away. â€Å"You didn't get light-headed or anything?† Jody asked. â€Å"I was a little hungover when I woke up, but I was fine after a couple of cups of coffee.† â€Å"Fuck!† Jody spat. Then she held her head. â€Å"Calm,† Tommy said, patting her shoulder. â€Å"Dr. Flood will make it all better. Maybe.† Jody growled, just loud enough for Tommy to hear. â€Å"Ya know,† said William, when there was a break in the pedestrian traffic and he didn't have to concentrate on looking pathetic, â€Å"I'm flush for cash, but since you're in the Christmas spirit, I'd still go for a look at Red's hooters.† â€Å"Bite me, dirtbag,† Jody said as she rolled up on William. â€Å"Honey.† Tommy caught the back of her newly purchased red leather jacket, just in case. They'd never know if his idea was going to work if Jody snapped the bum's neck. â€Å"I will not be sexually harassed by the entree.† â€Å"Something you ate isn't agreeing with you?† Tommy grinned at her when she looked back at him, but the fire went out of her eyes. â€Å"You can just cross sweet monkey love right off your list,† Jody said. â€Å"Jeez, what a bitch,† said William. â€Å"Her time of the month?† Tommy quickly wrapped his arms around Jody, lifted her off her feet, and carried her a few steps around the corner, even as she squirmed. â€Å"Let me go, I'm not going to hurt him.† â€Å"Good.† â€Å"Much.† â€Å"That's what I thought,† Tommy said, still holding her tight. â€Å"Why don't you head over to the Walgreens and I'll finish up with the huge cat guy?† A family of Christmas shoppers smiled as they passed them, thinking they were young lovers indulging in a public display of affection. The father whispered â€Å"Get a room† under his breath to his wife, which a normal person wouldn't have heard. â€Å"Count your lucky stars, buddy, we almost did it in the Santa's Workshop window. Hot, sweaty elf sex – in front of the kids. The kids would have liked that, huh?† The father hurried his family on down the street. â€Å"Nice,† Jody said. â€Å"Way to stay under the radar.† â€Å"Well, you know, I like to stay sharp,† Tommy said. Because he was nineteen and had only started having sex regularly since he met Jody, he still thought he had some sort of secret knowledge that was unavailable to other people. How can they possibly be thinking about anything else? he thought in the private part of his mind. â€Å"I'll bet it smells like peppermint,† Tommy said. â€Å"What?† â€Å"Elf sex.† â€Å"Would you please put me down.† â€Å"Okay, but don't hurt the huge cat guy.† â€Å"I'm fine. I'll meet you at the drugstore in five minutes. This had better work.† â€Å"Five minutes,† Tommy said. â€Å"Cinnamon. Maybe it smells like cinnamon.† The pale couple stalked the aisles of the Walgreens, having a great time dismissing the crass accoutrements of bourgeois American life, and generally scoffing at all the conventions of traditional culture. They were elite, after all. Special. Chosen – if you will – if only by the nature of their heightened sensitivities and superior sensibilities. They both claimed the ability to look past the faà §ade put on by most people, and see the very depths of the human soul. Strange, then, that they didn't see it coming when the skinny guy in a flannel shirt popped around the corner in front of them. â€Å"Let's ask these guys,† Flannel said. â€Å"They look like heroin addicts.† Jared White Wolf and Abby Normal backpedaled from the eyeliner display where they'd been looking for something hypoallergenic. Abby's eyes had been watering all night, causing her makeup to run and giving her more of a sad-clown-of-life look than she was going for. Jared hid behind Abby, just a little, which was awkward, since he was nearly a foot taller than she. The guy in flannel was joined by a beautiful, pale redhead, carrying an armload of toiletries. What amazing hair, Abby thought, looking at the long red tresses. I'd give anything for hair like that. â€Å"Tommy, leave these poor people alone,† said the redhead. â€Å"No, wait.† Flannel turned to Abby and smiled. â€Å"Do you guys know where they keep the syringes?† Abby looked at Jared, who looked at the guy in flannel. â€Å"Well, you can't just buy them,† Jared said. He was fiddling with the leather straps on his bondage pants, looking coy. Abby slapped his hand. â€Å"You need a prescription to buy syringes,† Abby said. â€Å"Do you really think I look like a heroin addict?† Jared threw his bangs out of his face dramatically. His head was shaved except for his bangs, which reached to his chin, specifically so he could throw them out of his face dramatically. â€Å"I was, like, thinking that maybe I should bulk up. You know, eat and stuff, but – â€Å" â€Å"Well, thanks,† said Flannel Shirt. The redhead moved off down the aisle. â€Å"I was going to try some heroin, but if you can't buy needles, well, there you go. See you guys. Nice shirt, by the way.† Abby looked down at her T-shirt, black, of course, with the image of a poet taken from a nineteenth-century etching. â€Å"Like you even know who it is.† † ‘She walks in beauty, like the night, † quoted the flannel-shirt guy. He winked at her, then grinned. â€Å"Byron's a hero of mine. See ya.† He turned and started to walk away. Abby reached out and snagged his sleeve. â€Å"Hey, there are needle exchange programs all over town. They're listed in the Bay Guardian.† â€Å"Thanks,† said flannel. He turned and Abby grabbed him again. â€Å"We're going to be at the Glas Kat. There's a Goth club tonight. Five-hundred block of Fourth Street. I know a dealer there. You know, for your heroin.† The flannel-shirt guy nodded, and looked at Byron's picture on her shirt again, then at her face. Fucksocks. He's so looking at my streaking eye makeup. â€Å"Thanks, milady,† said Flannel Shirt. And he was gone, off over the dark moors of the tampon aisle. â€Å"What was that about?† whined Jared. â€Å"He's so, so Happy Days.† Jared White Wolf spent a lot of time watching Nick at Nite when he wasn't brooding or fussing with his appearance. Abby walked into the flap of Jared's black duster and pounded his slight chest with her palms. â€Å"Didn't you see. Didn't you see?† â€Å"What, you acting like a complete ho?† â€Å"He had fangs,† Abby said. â€Å"Well, so do I,† Jared said, reaching into his pocket and pulling out a pair of perfect, dentistry-quality vampire fangs. â€Å"Duh, everybody does.† â€Å"Yeah, but his grew! I saw them. Let's go,† Abby said, pulling Jared White Wolf by his great bat-wing lapels. â€Å"I have to change into something hot before we go to the club.† â€Å"Wait, I want to get some Halls. My throat is raw from all the cloves we smoked last night.† â€Å"Hurry.† The buckles of Abby's black platform boots jangled as she dragged her friend past the lipsticks and hair products before he could get interested. â€Å"Okay,† said Jared, â€Å"but if I don't meet a cute guy tonight, you have to stay up all night and hold me while I cry.† â€Å"You should try black lipstick sometime,† Tommy said to Jody as they approached their building, their arms loaded with packages. He was still thinking about the kids at the drugstore. It was the first time since tenth grade that he'd used his knowledge of Romantic poetry. For a while he'd tried molding himself into the tragic Romantic hero, brooding and staring clench-jawed off into space as he composed dark verse in his head. But it turned out that trying to appear tragic in Incontinence, Indiana, was redundant, and his mother kept shouting at him and making him forget his rhymes. â€Å"Tommy, if you keep grinding your teeth like that, they'll wear away and you'll have to have dentures like Aunt Ester.† Tommy only wished his beard was as heavy as Aunt Ester's – then he could stare out over the moors while he stroked it pensively. â€Å"Yeah,† Jody said, â€Å"because I need to make it more obvious that I'm an undead creature that feeds on the blood of the living.† â€Å"You make it sound so sordid.† â€Å"No, I meant it in a nice way.† â€Å"Oh.† â€Å"Because it's not like people wouldn't understand if they found out we were vampires, because we slipped up and, oh, I don't know, UNSHEATHED OUR FANGS IN THE FUCKING DRUGSTORE!† Tommy almost dropped his packages. She hadn't said a word about that all night. He'd hoped she hadn't noticed. â€Å"It was an accident.† â€Å"You called that girl ‘milady. â€Å" â€Å"She was impressed with my Byron.† â€Å"Yeah, well, your Byron was probably sticking out a little, too, wasn't he?† â€Å"It wasn't like that.† â€Å"You drooled.† Jody paused at their security door and dug into her jacket for her key. Tommy stepped around her. â€Å"I'm still new at this. I think I'm doing pretty well. My ghastly pallor obviously impressed the lady at the needle exchange.† He reached into his bag and fanned out a handful of sterile-wrapped and capped syringes. â€Å"Congratulations, you can now pass as an HIV-positive heroin addict.† â€Å"Trs chic.† He grinned like he imagined a sexy Italian man-whore might. â€Å"Who drools in public,† Jody said. Damn, she's immune to my sexy Italian man-whore grin, Tommy thought. He said, â€Å"Be nice, I'm new. My lips don't fit together right when my fangs are out.† She turned the key and swung the door open. There, passed out on the landing, was William the huge cat guy and sleeping on his chest, Chet the huge cat. â€Å"I told you it would work,† Tommy said. Jody stepped into the stairwell and closed the door behind her. â€Å"You go first.† Fifteen minutes later, as he placed five syringes full of blood in their refrigerator, Tommy said, â€Å"This vampire thing is going to be great.† He'd had a moment when he'd bitten William – not just getting over the idea of being that close someone who smelled that nasty, but also being close to another man period. But after cleaning William's neck with an alcohol swab they'd gotten from the drugstore, and consoling himself that most literary vampires seemed sexually ambivalent anyway, the blood hunger pushed him through. He was feeling more relaxed, now that they had the food problem solved – for a while, anyway. If his friends didn't kill them in the next couple of days, he might even enjoy life as a vampire. Then he turned to Jody and frowned. â€Å"But I can't help but think that it may be wrong, taking advantage of a homeless alcoholic.† â€Å"We could just hunt and kill people,† Jody said cheerfully. She had a little crust of William's blood in the corner of her mouth. Tommy licked his thumb and wiped it away. â€Å"We did give him a nice sweater for his huge shaved cat,† Tommy said. â€Å"I loved that sweater,† Jody said. â€Å"And we are giving him a warm landing to sleep on,† she added, diving onto Tommy's rationalization dog pile. â€Å"And if we only take a little bit each day, he'll actually feel better. I know I did.† â€Å"And we won't become alcoholics ourselves.† â€Å"How are you feeling, by the way?† Tommy said. â€Å"Better. Hair of the dog. You?† â€Å"Two-beer buzz, max. I'll be fine. You want to try the experiment?† Jody checked her watch. â€Å"No time. We'll do it tomorrow night.† â€Å"Right. So, on to the list. Looks like hot monkey love.† â€Å"Tommy, we need to find a daytime person to help us. We have to move out of this place.† â€Å"I've been thinking about Alaska.† â€Å"Okay, good for you, but we still need to find a place to live where the Animals and Inspector Rivera can't find us.† â€Å"No, I'm thinking we should move to Alaska. For one thing, in the winter, it's dark for like twenty hours a day, so we'd have plenty of time. And I read somewhere that Eskimos put their old people out on the ice when they are ready to die. It would be like people were leaving snacks out for us.† â€Å"You're kidding.† â€Å"Eskimo Pies?† He grinned. Jody put her hand on her hip and looked at him, her mouth hanging open a little, as if she was waiting for something more. When it didn't come, she said, â€Å"Okay then, I'm going to change.† â€Å"Into a wolf?† â€Å"Clothes, cadaver breath.† â€Å"I didn't know. I thought maybe you'd learned.† Tommy thought Alaska was a great idea. Just because she was a few years older, she always acted like his ideas were stupid. â€Å"The thing with William worked,† he said defensively as he put away the supplies they'd bought at the drugstore. â€Å"That was a good idea,† Jody said from inside the closet. Now what? â€Å"Well, Alaska isn't a bad idea.† â€Å"Tommy, there's like nine people in all of Alaska. We'll stand out, don't you think?† â€Å"No, everyone is pale there. They don't have sun for most of the year.† She came out of the closet wearing her little black cocktail dress and her strappy come-fuck-me pumps. â€Å"I'm ready,† she said. â€Å"Wow,† Tommy said. He'd forgotten what they were talking about. â€Å"You think the Ferrari-red lipstick would be too much?† â€Å"No, I love the Ferrari-red lipstick on you.† Hot, sweet monkey love, he thought. This was exactly why he loved her. In the midst of all of the pressure, the danger, really – she still took time to think of his feelings. She lifted her breasts until they threatened to spill out of the plunging neckline of the dress. â€Å"Too much?† â€Å"Perfect,† Tommy said, walking toward her with his hands out. â€Å"Gimme.† She breezed past him into the bathroom. â€Å"Not for you. I need to get going.† â€Å"No, no, no,† Tommy said. â€Å"Hot monkey love.† While Tommy watched from the doorway, Jody applied the Ferrari-red lipstick, checked it, then frowned and wiped it off, then grabbed a different tube off the vanity. â€Å"When I get back.† â€Å"Where?† Tommy said. Sexual frustration had reduced him to single syllables. She turned to him with the new coat of maroon lipstick. â€Å"To get your minion.† â€Å"Not like that, you're not,† Tommy said. â€Å"This is how it works, Tommy. Respiratory Syncytial Virus (RSV) Vaccine Formulation Abstract Respiratory syncytial virus (RSV) is the leading cause of upper and lower respiratory tract infections in both children and elderly people. In this study we evaluated the short- and long-term protective efficacy of the single intranasal immunization of RSV vaccine formulation consisting of the fusion (F) protein formulated with a polyIC, an innate defense regulator peptide and a polyphosphazene (ΆF/TriAdj). This vaccine induced significant systemic and local immunity, including RSV F-specific IgG1 and IgG2a, IgA and virus neutralizing antibodies, and IFN-ÃŽÂ ³-secreting T cells in mice. Furthermore, this vaccination promoted the induction of RSV F85-93-specific CD8+ effector T cells with no virus replication in the lungs. To evaluate the duration of immunity induced by single intranasal vaccination, mice were challenged with RSV after five months of immunization. Significantly higher levels of IgG1, IgG2a and virus neutralizing antibodies were detected in ΆF/Tr iAdj vaccinated animals. Moreover, this vaccine formulation induced high local IgA secreting memory B cell development and B cell IgA production and conferred complete protection against subsequent RSV challenge without priming for enhanced disease. In conclusion, a single nasal vaccination of RSV ΆF protein formulated with TriAdj enhanced robust, long- term protective immune responses against RSV infection. Introduction Respiratory syncytial virus (RSV) is a major cause of severe lower respiratory tract disease in infants, older and immunocompromised individuals worldwide {Falsey, 2005 #586}. Many RSV vaccine candidates, including live attenuated strains, vectored and viral protein subunit vaccines are underway. Currently, there is no licensed RSV vaccine or specific treatments other than prophylaxis with passive antibody therapy (Palivizumab) {Group, 1998 #599}. The first RSV vaccine clinical trial with formalin-inactivated RSV (FI-RSV) failed to induce neutralizing antibodies and caused enhanced respiratory disease after natural RSV infection {Kim, 1969 #589}. RSV is an enveloped virus, containing three transmembrane surface glycoproteins: the major attachment protein G, fusion protein F and small hydrophobic SH protein. The F protein is highly conserved and mediates fusion of the virus into host cells and subsequent formation of syncytia, making it a major target for subunit vaccine and antiviral drug development {Collins, 2008 #597}. However, mucosal vaccination with RSV F protein alone does not induce protective immunity {Vaux-Peretz, 1992 #648;Garlapati, 2012 #650} therefore, this protein needs to be combined with an adjuvant to induce strong, long-lasting protective immune responses. Previously, we developed a novel combination adjuvant platform (TriAdj) comprising of three components, namely a polyI:C, innate defense regulator peptide (IDRs) and polyphosphazene (PCEP). Synthetic double-stranded RNA, polyIC, known to recognize TLR3 and retinoic acid-inducible gene 1(RIG-I) leading to induction of proinflammatory cytokines, which in turn activates various immune cells {Sivori, 2004 #581`, Longhi`, 2009 `#573;Longhi, 2009 #573}. To stabilize and protect the polyIC from degradation, two other immunmodulators an innate defense regulator (IDR) peptide and a polyphosphazene were added. Innate defense regulator (IDR) peptides are derivatives of natural host defense peptides with microbicidal, chemotactic properties {Yeung, 2011 #608}. Poly[di(sodium carboxylatoethylphenoxy)]-phosphazene (PCEP) is a synthetic biodegradable water-soluble polymer with immunostimulatory properties. It forms non-covalent complexes with antigens and/or other adjuvants to increase their stabil ity and allow multimeric presentation {Andrianov, 2009 #618;Kovacs-Nolan, 2009 #621}. Previously, we developed a novel vaccine candidate against RSV (ΆF/TriAdj) consisting of a truncated version of the RSV fusion protein (ΆF) formulated with TriAdj. We have shown that double vaccination regime such as prime and boost intranasal immunization with this vaccine candidate establishes long-lasting humoral and cell-mediated protective immune responses without any evidence of immunopathology {Garg, 2014 #60;Garg, 2015 #63}{Garg, 2014 #60}{Garg, 2014 #60}{Garg, 2014 #60}{Garg, 2014 #60}{Garg, 2014 #60}. In the present study we demonstrated that after single IN delivery with ΆF/TriAdj, vaccine elicits mucosal and systemic immune responses and offers complete protection from RSV challenge in mice. Furthermore, ΆF/TriAdj induced effective local CD8+ T cells, which is one of the hallmarks for successful vaccination against many viral infections. To evaluate the duration of immunity induced by this vaccine candidate after single intranasal vaccination, we carried out long-term trials. Five month after the vaccination, RSV ΆF formulated with TriAdj induced robust mucosal and systemic immune responses and complete protection from RSV challenge in mice. Furthermore, ΆF/TriAdj elicited long-lived local IgA secreting memory B cell development and B cell IgA production, as well as memory T cells. It is well known that an RSV vaccine needs to induce a balanced Th1/Th2 adaptive immune response, which stimulates the generation of high-affinity neutralizing antibodies with effector cytotoxic CD8+ T cells {Graham, 2011 #642}. Results Single mucosal vaccination with ΆF/TriAdj induces strong systemic immune responses: Antibodies play a major role in protection against many viruses. To characterize the quality of the humoral immune responses induced by single vaccination of ΆF/TriAdj, IgG1 and IgG2a titers were measured in the serum. As shown in Fig. 1A B, the mice immunized with ΆF/TriAdj developed significantly higher IgG1 and IgG2a levels, than placebo groups both before and after challenge with RSV. These results indicate that formulation of ΆF protein with TriAdj promotes a balanced humoral immune response. Neutralizing antibody plays a protective role against viral infections. In order to evaluate the biological function of the ΆF-specific serum antibodies, Virus neutralization (VN) titers were determined. Mice immunized with ΆF/TriAdj developed significantly higher neutralizing antibody titers than the ones immunized with PBS both before and after challenge with RSV (Fig 1C). To investigate the phenotype of the cellular immune response, the ΆF-induced secretion of IFN-ÃŽÂ ³ and IL-5 by splenocytes was measured four days after challenge. Significantly higher numbers of IFN-ÃŽÂ ³ secreting cells were generated from mice vaccinated with ΆF/TriAdj formulation, while the number of IL-5 secreting cells was low in vaccinated mice (Fig 1D). This suggests the addition of RSVΆF protein to the TriAdj resulted in a stronger, Th1-biased immune response, which is in also agreement with the enhanced IgG2a production. Intranasal vaccination with single dose of ΆF protein formulated with TriAdj induces robust protective mucosal immune: Mucosal immune response play a crucial role in first line defense of host by blocking the spreading of RSV infection in the lungs. Thus, an effective RSV vaccine should induce local IgA antibodies in the respiratory tract. To evaluate the mucosal immune responses induced by ΆF/TriAdj, we measured ΆF-specific IgA antibody levels in the lungs. Significantly higher IgA production was observed in mice immunized with ΆF/TriAdj in comparison with mice immunized with PBS (Fig 2A). To determine whether single IN vaccination with the ΆF/Triadj formulations would affect viral clearance, all mice except those in one of the two Placebo groups were challenged IN with RSV. No infectious virus was recovered in lungs of mice immunized with ΆF/TriAdj, showing full protection from infection (Fig 2B). In order to investigate whether protection is correlated with higher cross-presentation and cell-mediated immune responses, we evaluated RSV F-specific CD8+ T cells infiltration with RSV F85-93 pentamers in the lungs after RSV challenge. The ΆF/TriAdj induced a significantly higher number of F85-93 specific CD8+ T cells influx in the lungs (Fig 6A), which suggests that vaccination with ΆF/TriAdj promotes a cytolytic CD8+ T cell response to RSV infection. Virus-specific effector CD8+ T-cell response has been shown to play a critical role in RSV clearance {Graham, 2011 #642}. Therefore, we analyzed the effecter function of the CD8+ T cells according to their expression of the IFN-ÃŽÂ ³ by à ¯Ã‚ ¬Ã¢â‚¬Å¡ow cytometry. After RSV challenge, the ΆF/TriAdj-immunized group showed higher frequencies of IFN-ÃŽÂ ³-expressing CD8+ T cells than PBS-immunized, RSV-challenged mice in the lungs (Fig 6). This suggests that ΆF/TriAdj promotes a stronger ef fector CD8+ T cell response to RSV. Single nasal vaccination with ΆF/TriAdj induces Long-term systemic immune responses: To assess the long term memory effect induced by single intranasal vaccination with ΆF/TriAdj, serum IgG and VN antibody levels were measured periodically over five months. The mice immunized with ΆF/TriAdj developed strong IgG and VN titers compare to placebo control group for approximately 25 weeks (Fig 3A B). We next examined whether IN vaccination promoted the induction of long term ΆF-specific serum IgG1 and IgG2a. The ΆF/TriAdj- vaccinated mice developed significantly higher ΆF-specific serum IgG1 and IgG2a titers than placebo groups both before and after challenge with RSV (Fig 4A B). VN titers were also determined to evaluate the functional quality of the ΆF-specific serum antibodies after RSV challenge. Mice immunized with ΆF/TriAdj developed significantly higher neutralizing antibody titers compared to PBS gro up (Fig 4C). To further define the phonotype of the immune response, the ΆF-induced secretion of IFN-ÃŽÂ ³ and IL-5 by splenocytes was measured four days after challenge with RSV. The ΆF/TriAdj formulation generated a significantly higher frequency of IFN-ÃŽÂ ³ secreting cells compared to the placebo. Mice immunized with ΆF/TriAdj developed significantly higher IgA levels when compared to placebo and placebo virus challenged groups (Fig 5A). Furthermore, we checked the development of IgA-secreting memory B cells in lungs after RSV challenge. ΆF/TriAdj formulations generated a significantly higher frequency of RSV ΆF-specific IgA-secreting memory B cells in lungs in comparison to PBS (Fig 5B). In summary, single dose of ΆF/TriAdj via IN vaccination stimulated strong IgA-secreting memory B cell development and B cell IgA production. To determine viral clearance after single vaccination with ΆF/TriAdj, all mice except those in one of the two placebo groups were challenged IN with RSV on day 150. The mice were sacrificed after four days, and virus titers in the lungs were determined. No infectious virus particle was recovered from mice vaccinated with ΆF/TriAdj, showing that these mice still had sufficient immunity to be completely protected from challenge virus replication in the lungs (Fig 5C). Discussion RSV is one of the major global burdens of causing a broad spectrum of respiratory illnesses in children and older population worldwide. There is still no licensed RSV vaccine, in part due to the disastrous outcomes observed following vaccination of naÃÆ' ¯ve children with FI-RSV {Kim, 1969 #589}. Natural infection with RSV fails to protect against subsequent infection because it stimulate modest immunogenicity and short-lived immunological memory against virus. Similar to natural RSV infection, many promising RSV vaccine have failed to generate long-lasting, protective immune responses {Power, 2008 #769;Hall, 2001 #789}. For wide clinical application, RSV vaccine will need to be safe, easy to administer and must stimulate strong long-term protective immunity than natural RSV infection {Pulendran, 2011 #796}. Protein subunit vaccines have a high safety profile, but are generally poorly immunogenic and induce short-lived humoral and cellular immunity {Graham, 2011 #642}. So adjuvant s are added to subunit vaccines to stimulate faster, stronger, and long-lasting immune responses to vaccines. At the clinical level, multi dose vaccination is expensive and has potential safety problems. Whereas, single vaccination formats are cost effective, safe (no risk for vaccine contamination) and more convenient to health worker. In the present study, we demonstrated the short as well as long term protective efficacy including stable systemic IgG, local IgA production and neutralizing antibody titers of a RSV ΆF protein formulated with TriAdj in mice after single IN immunization. Furthermore, ΆF/TriAdj promoted an effector CD8+ T cells with compete protection against RSV infection without inducing vaccine-enhanced pathology in the lungs. A vaccine requiring only a single administration to be effective would be highly practical, as it would improve patient compliance Therefore, single intranasal immunization with RSV ΆF protein formulated with Tri Adj is a promising approach to induce a long-lasting protective RSV-specific immune response. Mucosal immunization via IN route is invasive and suitable for generation of both local and systemic immunity. RSV infect through respiratory tract, therefore mucosal antibodies specifically IgA play an important role in prevention and clearance of virus. Previously it has been shown that protection against RSV infection is more correlated to the levels of mucosal IgA rather than to systemic antibody in human {Watt, 1990 #687}. In mice, single RSV infection induces short-lived neutralizing antibodies with absence of antibody-secreting memory B cells {Schmidt, 2012 #869}. Interestingly, we have recently demonstrated that the RSV infected mice were showed significantly lower level of local B cell IgA production and IgA secreting memory B cell development that indicates an impaired local antibody response that allows RSV re-infection and explain the short-term protective immunity with natural infection with RSV{Garg, 2016 #1340}. In contrast, intranasal vaccination with single dose of R SV ΆF protein formulated with TriAdj promotes the induction of long-lasting local IgA secreting memory B cell development and B cell IgA production which is a major criterion for an effective RSV vaccine. Previously, we and others have reported that strong, effective and protective immune responses were induced only after a booster immunization with RSV F protein with adjuvant {Garg, 2014 #891}{Blanco, 2014 #1423}{Lambert, 2015 #1442}. A single mucosal vaccination of adenovirus-based RSV vaccine had also been proved effective in short term {Kim, 2010 #1443}. But this is the first report showing the long term protective efficacy of single mucosal vaccination of an adjuvanted RSV F protein subunit vaccine in mice. Clinical trial with FI-RSV failed to induce protection against RSV due to generation of poor neutralizing antibodies and TLR activation that led to vaccine-associated enhanced disease {Delgado, 2009 #513}. The inclusion of polyI:C as TLR ligand in vaccine formulation, is expected to overcome the limitations of inactivated RSV vaccines. The adjuvant effect such as long-lasting T cell immunity of polyI:C is likely caused by the direct interaction with PRRs such as TLR3, MDA5 and RIG-I, leading to production of pro-inflammatory cytokines, and chemokines. Interestingly, it has been shown that alveolar macrophages and lung DCs induced strong IgA and IgG antibodies by addition of TLR3 ligands, which were probably linked to secretion of BAFF/APRIL cytokines and activated B cells {Xu, 2008 #686}. However, we and others have been shown that IN administration of higher doses of polyIC caused marked production of inflammatory cytokines/chemokines accompanied by impaired lung function {Stowell, 2009 #682;Boukhvalova, 2010 #685;Aeffner, 2011 #750} suggesting excessive stimulation of local immune responses can result in detrimental effects. In contrast, ΆF/TriAdj has an advantage over polyIC alone as it has shown full protection form RSV infection without any exacerbation of chronic pulmonary disease. This demonstrates that a combination of polyIC, IDR and PCEP mediates optimal enhancement of the RSV-specific immune response without vaccine enhanced disease. We believe that IDRs is one of the major anti-inflammatory components in our vaccine formulation as it has been previously shown to enhance cell-mediated immune responses and to modulate excessive consequences of TLR signaling {Bowdish, 2005 #766;Hancock, 2006 #767}, whereas polyphosphazenes enhance antigen-specific humoral immunity through the formation of non-covalent complexes with protein {Andrianov, 2005 #667}, suggesting major roles from both of these compounds in this vaccine formulation. An important factor for successful vaccine against RSV requires the generation of effective CD8+ T cell in the lungs. Previously, it has been shown that mild to severe infection with RSV failed to induce virus-specific IFN-ÃŽÂ ³ recall responses in the infants {Lee, 2007 #806}. To define the stimulation of local effective CD8+ T cell responses induced by ΆF/TriAdj, RSV F specific pentamer staining demonstrated marked induction of IFN-ÃŽÂ ³ secreting RSV-specific CD8+ T cells in lungs. Similarly, exogenous IFN-ÃŽÂ ³ expression protects against RSV infection in the lungs of BALB/c mice {Kumar, 1999 #805}. This evidence suggests that complete protection against RSV was associated with the presence of IFN-ÃŽÂ ³+ RSV-specific CD8+ T cells. In summary, our data show that RSV ΆF protein formulated with TriAdj vaccine represents a safe, effective and promising RSV vaccine candidate. The major advantages associated with this mucosal RSV vaccine is to induction of long-term protective immunity with a single mucosal vaccination by stimulating long-lived RSV-specific neutralizing antibodies, memory B and CD8+ T cells, therefore warranting additional evaluation as a vaccine against RSV in clinical trials. MATERIALS AND METHODS Virus and vaccine formulation: The RSV (A2 strain) was propagated in Hep-2 cells (American Type Culture Collection, VR-1540). The RSV ΆF protein with his-tag was produced and purified as described previously {Garlapati, 2012 #650}. Briefly, HEK-293T cells were transfected with an episomal vector expressing the ΆF protein using Turbofect (Fermentas, R0534). The ΆF protein with his-tag was purified using TALON Superflow resin (Clontech,) according to the manufacturers instructions. The ΆF protein was formulated with 10  µg poly (I:C) (Invivogen, tlrl-picw), 20  µg IDR1002 (Genscript, 818360) and 10  µg PCEP (Idaho National Laboratory) in PBS (ΆF/TriAdj) as described previously. Animals, immunizations and challenge: Six to eight week old female BALB/c mice (Charles River Laboratories, Wilmington, MA, USA) were immunized once IN with ΆF/TriAdj. Two additional groups of mice received PBS IN (Placebo control). Three week (short-term trial) and twenty weeks (long term trial) post-vaccination, all except one of the Placebo groups were challenged IN with RSV strain A2 (5 x 105 PFU/50 µl). Mice were euthanized four days after RSV challenge for detection of immune response and virus in the lungs. Blood samples were drawn by cardiac puncture under anesthesia prior to challenge and at regular intervals afterwards. All procedures were approved by the University Animal Ethics Committee in accordance with the standards stipulated by the Canadian Council on Animal Care. Lung fragment cultures and ELISA: Lavaged lungs of euthanized mice were cut into small pieces and lung fragment cultures were prepared as described previously {Mapletoft, 2008 #591}. ELISA was performed as described previously {Garg, 2014 #942}. RSV ΆF-specific IgG1, IgG2a and IgA were detected using biotin-labeled goat anti-mouse IgG1, IgG2a (Southern Biotech, 1070-08, 1080-08) or IgA (Gibco, M3115) followed by streptavidin-alkaline phosphatase (AP) (Jackson ImmunoResearch Laboratories Inc.,016-050-084), and developed with p-nitrophenyl phosphate (Sigma-Aldrich, N3254) substrate. Enzyme-linked immunospot (ELISPOT) assays: ELISPOT assays were performed on splenocytes and LNs cells as previously described {Garlapati, 2012 #650}. Briefly, cells were stimulated with ΆF protein (2  µg/ml) and spots were developed using biotinylated IFN-ÃŽÂ ³- or IL-5- (BD Biosciences, 554410, 554397) or goat anti-mouse IgA specific antibody, AP-conjugated streptavidin and BCIP/NBT (Sigma-Aldrich, B5655) as the substrate. Analysis of CD8+ T cells from lungs by flow cytometry: Lung single-cell suspensions were generated as previously described {Garg, 2014 #942}. To analysis of RSV F85-93-specific CD8+ T cells, lung mononuclear cells were surface stained with H-2Kd-F85-93 MHC Class I pentamer (ProImmune, F149) together with labeled antibodies specific for CD8+ T cells (BD Pharmingen, 553031). For intracellular detection of IFN-ÃŽÂ ³ in CD8+ T cells, lung mononuclear cells were stimulated with 1 ÃŽÂ ¼M of F85-93 peptide (KYKNAVTEL) and cell surface staining was performed, followed by intracellular cytokine staining using Cytofix/Cytoperm (BD Pharmingen, 554715) and APC- conjugated anti-IFN-ÃŽÂ ³ antibody (BD Biosciences, 554413). After staining, cells were acquired by flow cytometry (BD Biosciences), and data were analyzed using Kaluza Software (Version 1.2). Cells were gated for live cells, singlets and lymphocytes and then analyzed for indicated markers. Virus titration and virus neutralization assay: Virus titrations were performed with individual lungs at day 4 post- challenge as described previously {Garg, 2014 #942}. Results are expressed as PFU/g of lung tissue. RSV-specific neutralization titers were determined by plaque reduction assays. Serum samples were mixed with 500 PFU/well of RSV strain A2 for 1 h at 37 °C. The sample-virus mixtures were transferred to HEp-2 cell monolayers and incubated for 4 days at 37  °C, and the cells were fixed and stained with 0.5% crystal violet. Statistical analysis: All data were analyzed using GraphPad PRISM version 6 for Windows (GraphPad Software, Inc., La Jolla, CA, USA). Differences among all groups were examined using Student t-tests, one-way ANOVA, followed by a Newman-Keuls post test. Virtual Reality Imagine walking down a white sandy beach. The sand is warm between your toes and there is a light breeze that moves your hair. You look across the horizon at the bright warm sun, a little to bright you think as you squint. But the water before you is so clear, you can see small fish clear to the bottom. The water is cool and it feels refreshing as you let the waves crash gently at your feet. This place is beautiful and peaceful, but you realize your not alone. You look to your right and see a child and her mother splashing in the water. You can hear the child laughing and feel the rippling sensation in the water against your legs. The sun is getting a little warm as a small bead of sweat falls across your brow. You look onto the beach and see the white sand as it glistens from the sun. You look a little farther and see the towel and chair you left earlier. You marvel about what a beautiful day it is. Then, you sigh as you walk across the warm sand and it sticks to your wet feet, wate r is dripping off you, but you feel relaxed. You don't want to leave this beautiful place, but you realize your time is up. You feel a sharp tap on your shoulder and your helmet is slowly being removed from your head. Reality comes flooding back to you, gone is the white sandy beach and warm sun, but and in its place are the cold uninviting walls of the laboratory. The scientist smiles and asks how you enjoyed your experience. What has just happened is the technology of the future called virtual reality. This is concept that allows one to think they are experiencing something through the use of the major senses. Virtual reality creates an imaginary image that allows for a sense of deception. Virtual reality takes place in many ... ...of funds. With further research and advancements comes the opportunity to entertain, train, as well as help people. Wouldn't we all like to experience a little vacation without the hassle of packing, flying or driving, and unpacking when we return. The Forgotten Female in the Works of Hemingway      Ã‚  Ã‚   Ernest Hemingway has often been accused of misogyny in his treatment of female characters (and, perhaps, in his treatment of women in his own life). "It is not fashionable these days to praise the work of Ernest Hemingway," says Frederick Busch. "His women too often seem to be projections of male needfulness" (1). Many of his stories are seen as prototypical bildungsroman stories--stories, usually, of young men coming of age. There are few, if any, stories in the canon of women coming of age, however, and Hemingway is not the first to suffer the wrath of feminist critics. But is this wrath justified?    In his dissertation, Mark G. Newton reviews some of the critical literature that places Hemingway within the misogynist genre. "Cliches [sic] abound," he says. "Hemingway was in search of his manhood (an ignoble quest?); he hated women; he had a "death wish" and a "thin persona"; he was the archpriest of violence, etc." (6). However, Newton sees women in Hemingway's works as the "positive life-directed force which transports the male Hemingway hero away from a debilitating wound" (2), and he places them into "[t]he roles manifested by Hemingway's women in aiding the hero": "Ideal Women," "Sister Guides," "Icons and Dream Visions," "Wicked Women Who Also Serve," "Feminine Points of View," and "Full Cycle." My problem with Newton's approach to the feminine in Hemingway is that Newton seems to accept that, in presenting women as archetypal Eve's, the woman as "help-meet"-type image, that Hemingway is somehow presenting women favorably.    A somewhat similar view is presented by Jeryl J. Regardless of race, country, or culture, people follow similar patterns of exploration, inventiveness, and creativity. And it is these tendencies which guide our development and this one of the reasons why are important for the humankind. Tendencies serve to help human beings survive and develop fully making something of them. Animals have instinct and are born complete with everything they need to survive in an environment. The human beings have tendencies which drive them to explore, develop and adapt in order to survive whatever the environment is. Human beings have both physical and psychological needs. The physical needs for food, shelter and clothing, and the psychological need for security, love, art, music and culture. It is the tendencies that drive the humankind to look for and create those things that meet his physical and psychological needs from within his environment. Montessori’s belief in the potential of every human being is grounded in the idea that the world can be changed for the better if education is adapted to he child’s true nature instead of being limited to the transmission of academic knowledge. However we also need to recognise that as members of the human species, children are born with certain genetic traits or potentialities, unique to human beings, Montessori called these traits, human tendencies. These tendencies relate directly to human needs: for example o ur need for food is linked with our tendency to explore. 1. Tendency for Exploration It is through movement that exploration began. People learn from the discoveries they make or that which others before them made. They look around and make connections with things in their environment also by smelling and hearing. The child will have the natural urge to explore their environment they are in and are constantly learning from what they see and hear around them. The child is learning how to use their basic senses as in see, taste, touch, hear and manipulate something or the other. In the first few months the child is learning how to move his arms and legs he is not very strong at this time his muscles are not developed yet and he just observes his surroundings. They imitate the actions of those around them. All the connexions from the brain at the birth, which has one hundred billion neurons, are not yet made; therefore they are waiting for being connected in the new environment. Even though we have an immense responsibility to provide the right environment because the child will trust the environment and he will trust him to do things by himself. The child is an explorer. He needs to explore the world on a level that he is capable of learning from. The function of the Assistant to Infancy is to put the child in touch with the many aspects of the world around him which will help him classify his impression, build new skills and knowledge. The world is an exciting place and the entire more so if the child is able to make his own discoveries about it. The Assistant will choose activities which may nurture his interest but be careful to help the child to point the discovery without actually making the discovery for him. That is all about the exploration. 2. Tendency for Order The human being must have order in his environment to be able to find his way back home after exploring his environment. The child requires from of order whenever he is exploring. This will help him in on many levels- if he wishes to carry out an activity successfully then order is required. Order and its extension into exactness and precision will help him to gain clear and accurate impressions so that he may understand his world better. External order can help create a logical and orderly mind. We can observe in the small babies their love for order in their surroundings. They cannot practically live in disorder, they are so aware of it, thing which is not happen with the grown-up children. They will expect to find objects where they first perceived them and will go great lengths to put them back if they are ‘out of place’. 3. Tendency for Orientation Through order we can build our orientation within our surroundings. The child needs some points of reference in the environment to orientate him and find things. And this means he can orient himself in his environment and to act with purpose. The more the child is helped in this area the more independent he can be. The child is able to interact, express thoughts and ideas with people in his environment through language. Prior to developing language, the baby only could communicate by crying which meant the adult had to guess his needs. With language, the child is better clearly able to express his needs and thoughts and become more independent and secure. 5.
